The stability and bioavailability of an active pharmaceutical ingredient (API) in a drug is key to that drug achieving its desired effect. Supersaturating drug delivery systems, such as amorphous solid dispersions, is one approach to improve solubility.
In this webinar, the speaker discusses increasing the aqueous solubility of orally administered poorly water-soluble drugs. This is done by creating a supersaturation state in the gastrointestinal lumen, thus improving bioavailability.
The speaker compares the technologies of hot melt extrusion (HME) and spray drying as two ways to get to a stable formulation. The discussion includes case studies of real-life pharmaceutical applications.
Speaker: Maria Inês Ré, director at Gala Platform, a branch of the engineering school École des Mines d'Albi in Albi, France.
Duration: 60 minutes
The search for effective ways to increase the solubility of drugs is constant.
Hydroxypropyl methylcellulose acetate succinate (HPMCAS) can keep poorly water-soluble drugs in a supersaturated state after dissolution, so it's become a more popular carrier for amorphous solid dispersions (ASDs). Yet HPMCAS has mostly been limited to ASDs prepared by spray drying. It would be preferable to use the solvent-free method of hot-melt extrusion (HME) to prepare ASDs, but the high temperatures required can degrade the polymer.
This webinar reviews the use of solid surfactants to reduce complex viscosity (and thus the processing temperature) of HPMCAS-based ASDs. The speaker presents a study where surfactant-drug-polymer mixtures were successfully extruded, and the resulting ASDs were developed into tablets. The analysis paves the way for greater use of HME in producing HPMCAS-based ASDs.
Speaker: Dr. Abu Serajuddin, PhD, Professor of Industrial Pharmacy at St. John's University in Queens, New York, USA.

Increasing absorption is one way for drug formulators to improve the performance of medicines. This webinar explores how to do just that working with solid dispersions as a way to improve absorption by enhancing a drug’s solubility.
Watch this webinar to learn more about dispersions produced using hot melt extrusion (HME) with polyvinyl alcohol (PVA) as the matrix polymer. The material will take a top-down molecular look at one-phase and two-phase solid dispersions and further delve into the performance of the two-phase solid dispersion.
Speaker: Dr. Thomas Kipping, Head of Drug Carriers at Solid Formulations R&D at Merck KGaA in Darmstadt, Germany.
