High-quality controls play an integral role in the successful development of CRISPR-Cas9 experiments, as well as the performance of high throughput CRISPR-Cas9 screens. Thermo Fisher Scientific provides a complete collection of validated positive and negative controls to help optimize delivery conditions, maximize editing efficiency, and establish hit selection criteria. These controls will help build robust assays with a better signal to noise ratio.

CRISPR positive controls | A CRISPR positive control is necessary to monitor the gene editing efficiency of the CRISPR reagents being used in the experimental design. Positive controls are validated gRNA sequences that have demonstrated high editing efficiency across different cell types. Some positive controls offered By Thermo Fisher Scientific offer up to 90% editing efficiency or greater in several cell types. Individual LentiArray and TrueGuide gRNAs are available. | Positive controls are intended to be used during assay development to determine the conditions that provide maximum editing efficiency in your cell models. Individual gRNAs against specific genes are available to serve as biological controls for both assay development and as on-plate positive controls when performing your screen.

In the following experiment, HT1080 cells were infected with LentiArray CRISPR Control Lentivirus, human HPRT, with GFP. This technique provides a visual readout of transduction efficiency and helps determine the multiplicity of infection (MOI) for the cell line being used. This example illustrates how green fluorescent protein (GFP) enabled controls can be used to optimize transduction and assay design.
