The SelectScreen P450 Profiling Service utilizes Thermo Fisher Scientific's well-established P450 BACULOSOMES reagents and biochemical Vivid assay platform to help overcome drug discovery bottlenecks. It offers a flexible approach to profiling, enabling rapid analysis of your compounds against a panel of P450s (1A2, 2C9, 2C19, 2D6, 3A4) with IC50 determinations performed in duplicate using 10-point titrations of your test compound.
A stringent validation process ensures the highest quality data possible for the 10-point titration performed in duplicate. An additional two control wells for each compound concentration tested are conducted to detect autofluorescent compound interference. Strict quality control protocols ensure that any assay results not meeting set specifications will be automatically repeated, this includes performing IC50s with a known inhibitor for each P450 on a per plate basis to ensure the validity of the data.
Listed below are the Isozymes (CYP450 and Vivid Substrate) available for profiling. Invitrogen is the original manufacturer of all P450 BACULOSOMES reagents used in the SelectScreen P450 Profiling Service; we ensure the consistency and reliability of each P450. Cytochrome P450 BACULOSOMES reagents are microsomes, prepared from insect cells infected with recombinant baculovirus containing a human P450 isozyme and a rabbit NAPDH-P450 reductase. P450 BACULOSOMES reagents offer a distinct advantage over human liver microsomes (HLMs). Only a single P450 isozyme is expressed, which ensures that your metabolism profile can be attributed to that isozyme, and not to other P450s or other drug metabolizing enzymes.
