Invitrogen Alexa Fluor 561 dye is a bright, yellow laser excitable dye that is optimized for flow cytometers with spectral capabilities. This fluorophore can be used for cell surface, intracellular, and intranuclear antigens. It should be used in complex multicolor panels to fit between fluorophores FITC and PE. In simpler panels, Alexa Fluor 561 can replace PE to reduce emission spreading into PE tandem dyes.

Flow cytometry analysis of C57BL/6 mouse splenocytes using CD4-Alexa Fluor 561. C57BL/6 mouse splenocytes were stained with CD45 Monoclonal Antibody, Alexa Fluor 532, CD3e Monoclonal Antibody, PE-Cyanine7, CD45R (B220) Monoclonal Antibody, PE, CD8a Monoclonal Antibody, PE-eFluor 610, CD25 Monoclonal Antibody, FITC and 0.06 µg of CD4 Monoclonal Antibody, Alexa Fluor 561. CD45 positive leukocytes were used for analysis and gated on CD3 positive/CD45R (B220) negative cells (left). CD3e positive cells were further plotted as CD4 versus CD8a positive cells (center) and CD4 positive cells were analyzed for expression of CD25 (right). This data was collected on a 5-laser Cytek Aurora full spectral cytometer. As shown here, Alexa Fluor 561 is compatible with PE and PE tandems when proper panel design principles are applied.

Balb/c mouse splenocytes (left) and SJL mouse splenocytes (right) were stained with Anti-Mouse CD19 Monoclonal Antibody, FITC and a non-cross reactive, human-specific CD4 Monoclonal Antibody (RPA-T4), Alexa Fluor 561. As shown here, Alexa Fluor 561-conjugated antibodies non-specifically stain B cells in SJL mice (right) and outbred, Swiss Webster mice (data not shown). Non-specific staining was not observed in BALB/c mice (left) and C57BL/6 mice (data not shown).
