  • You can now use a sample input volume of 200 μL (vs. the standard 400 μL) with a reduced (halved) amount of extraction reagents.
  • This means that you can now get up to 2,000 reactions from your MVP I (A48310) or MVP II (A48383R) products. Additionally, the smaller pack size of the MVP I kit (A42352) can support up to 200 reactions.
What is the difference between the two kits?Binding Beads: In order to support the unprecedented demand for RNA extraction products for COVID-19 testing, there has been a raw material change in the bead component contained in the MagMAX Viral/Pathogen Nucleic Acid Isolation Kit. The MagMAX Viral/Pathogen II Nucleic Acid Isolation Kit contains these beads. The chemistry driving the RNA to the beads is identical, and the nucleic acid binding capacity of each bead type is sufficient to provide similar results between the two products.

Bulk reagent format: The MagMAX Viral/Pathogen II kit is only available in bulk prep format with 1,000 or 2,000 reactions.
The MVP Ultra kit includes a proprietary enzyme mix that enables lysis and nucleic acid recovery from the toughest pathogens, such as gram-positive bacteria, yeast, and fungi.

Sample input volume reduction

The following protocols can be used for either the MagMAX Viral/Pathogen Kit (MVP I), MagMAX Viral/Pathogen II Kit (MVP II) and MagMAX Viral/Pathogen Ultra Kit for extraction of SARS-CoV-2 and other infectious disease pathogens, which supports sample input volume from 5 mL to 200 uL and reduction from 3 wash steps to 2 wash steps. Saliva can be used to extract and analyze viral nucleic acid

Extract and analyze viral nucleic acid directly from a saliva sample. The following protocol can be used for either MagMAX Viral/Pathogen (MVP I) and MagMAX Viral/Pathogen II (MVP II) for extraction of SARS-CoV-2 and other infectious disease pathogens, which support the isolation of nucleic acid from 200 uL of saliva (raw and preserved).

Q. What is the main benefit of MVP Prime (A58145) compared to original MVP I kits (A42352, A48310), MVP II kit (A48383R) and the MVP Ultra Kit (A42356)

A. The MagMAX Prime Viral/Pathogen NA Isolation Kit offers several key benefits. Firstly, it utilizes our most advanced formulation, guaranteeing maximum nucleic acid yield and enhancing assay sensitivity. Additionally, it enables users to conveniently process various sample types on a single plate using the same KingFisher script. Lastly, this kit is compatible with the KingFisher Flex, Apex, Apex Dx (RUO mode), and Presto sample purification instruments, providing an automated workflow for seamless integration.

 

Q. What is the difference between the original MVP I kits (A42352, A48310) and the MVP II kit (A48383R)?

A. There are two key differences between the MVP I and MVP II products:

Binding Beads: A proprietary raw material change in the bead component is different. The chemistry driving the RNA to the beads is identical and the nucleic acid binding capacity provides similar/equivalent results between the two products. Bulk reagent format: The second difference is that the MVP II (A48383R) product is only available in the bulk format. There is no 100-reaction option for the MVP II kit.

 

Q. Can the MVP I, MVP II, and MVP Ultra kits be used without an instrument?

A. Yes, we have a manual ready protocol and user guide for use with a magnetic stand.

 

Q. Are other product contents and workflow the same for MVP I kits (A42352, A48310) and the MVP II kit (A48383R)?

A. Yes, the product contents (Binding Solution, Wash Buffer, Proteinase K, and Elution Solution formulations) are the same in both kits.

Yes, the overall workflow, in terms of number of steps and volume usage, remains the same; if you have been using the MVP I (A42353, A48310) kit you should not have to change your protocol.

IMPORTANT: The protocol for both the MVP I and MVP II products has been updated to include a separate lower input option: You can now use a sample input volume of 200 μL (vs. the standard 400 μL) with a reduced (halved) amount of extraction reagents.  What does this mean? It means that you can now get 2,000 preps from your MVP I (A48310) or MVP II (A48383R) products. This also means that the smaller pack size of the MVP I kit (A42352) can support 200 preps.

 

Q. Can I use the same KingFisher Flex script for both the MVP I and MVP II kit?

A. Yes, there will be a new script for the 200 μL sample input protocol and both kit versions can be used with it.

 

Q. Can I use the same samples with the MVP II kit as I am with the MVP I kit?

A. Yes, all the same samples are compatible.

 

Q. Is MVP II compatible with the TaqPath COVID-19 Combo Kit?

A. Yes. You can use the MVP II (A48383R) product just as you would use the MVP I (A42352, A48310) product with the TaqPath COVID-19 Combo Kit. When using the 200 μL sample input protocol, please note that the eluate input into the PCR will be changed to 10 uL (5uL eluate input when using 400 μL).

 

Q.  Is the new MVP II kit replacing the current MVP I kit?

A. No, the MVP II is NOT replacing the existing MagMAX Viral/Pathogen Kit (Cat. Nos. A423352, A48310).  The intent of this product to expand the number of samples that can be processed with our MagMAX reagents.

 

Q.  Are both the MVP I and MVP II products manufactured under the same ISO 13485 quality standards?

A. Yes, they are both manufactured under the same ISO 13485 standards which utilizes a high set of quality control measures.

 

Q:  Is there a difference in price?

A: No, the bulk reagents for both kits are the same price.

 

Q: What is volume input range for the kit?

A: For the 96DW plate format: 200-400 μL sample volume input (200 μL for whole blood).
For the 24DW plate format: 500-2000 μL sample volume input (1000 μL for whole blood).

 

Q: What sample types are compatible with the kit?

A: Human biofluid samples (e.g. blood, serum, plasma, urine, CSF, lavage, saliva), samples stored in transport media (e.g. BD® Universal Viral Transport media), and bacteriophage culture.

 

Q: What is the lower limit for viral titer?

A: Nucleic acid has been successfully detected from as low as 50 viral copies in the starting sample.

 

Q: What types of pathogens are compatible with the MVP I and MVP II kits?

A: (-)ssRNA virus, (+)ssRNA Virus, ssDNA virus, dsDNA virus, Gram negative bacteria.

 

Q: What minimum types of downstream analysis is the extracted nucleic acid compatible with?

A: qRT-PCR, qPCR, sequencing.

 

Q: How long is the protocol?

A: 30 min from sample to extracted nucleic acid for up to 96 samples using the MVP I and MVP II kits. 65 min from sample to extracted nucleic acid for up to 96 samples using the MVP Ultra kit.

 

Q: Is the kit automation compatible?

A: Kit is compatible with all KingFisher instruments (Flex, Duo Prime, and Presto) and can also be used for manual processing in 96-well plate.

 

Q: Which extraction kit is recommended with the qPCR TrueMark Plus Panels for respiratory, urinary, or sexually transmitted infection detection?

A: MagMAX Viral/Pathogen Ultra Nucleic Acid Isolation Kit (A42356) is recommended for all variations of the TrueMark panels, especially if a single extraction kit is preferred for a simplified workflow. This kit is especially effective for difficult to lyse samples containing gram-positive bacteria, yeast, or fungi pathogens.

Alternative workflows using MagMAX Viral/Pathogen I (MVP I) Nucleic Acid Isolation Kit (A42352, A48310) and MagMAX Viral/Pathogen (MVP II) Nucleic Acid Isolation Kit (A48383R) can also be used with the TrueMark panels.

 

Q: Which extraction kit is recommended with the qPCR, Applied Biosystems TrueMark STI Select Panel panels for sexually transmitted pathogen detection?

A: MagMAX Viral/Pathogen (MVP II) Nucleic Acid Isolation Kit (A48383R) is recommended for both feasibility and effectiveness to be utilized with the TrueMark STI Select Panels in detection of high-incidence sexually transmitted pathogens.

Alternatively, MagMAX Viral/Pathogen Ultra Nucleic Acid Isolation Kit (A42356) is also compatible with the TrueMark STI Select Panels.

Alternative workflows using MagMAX Viral/Pathogen I (MVP I) Nucleic Acid Isolation Kit (A42352, A48310) and MagMAX Viral/Pathogen (MVP II) Nucleic Acid Isolation Kit (A48383R) can also be used with the TrueMark panels.
