Gibco performance serum-free media (SFM) and animal origin-free (AOF) media deliver superior cell growth and virus production to serum-supplemented systems while maximizing consistency and reliability and simplifying downstream purification.
Gibco CD BHK-21 Production Medium is the first cell culture medium supporting complete serum-free and protein-free processes for BHK-21 cells. Because it does not require supplementation with serum or other undefined components, Gibco CD BHK-21 Production Medium eliminates the serum associated risks while allowing you to achieve better cell growth and more consistent viral yields.
Maintain high level of cell viability:
Without supplementation with FBS or TPB (Tryptose Phosphate Broth) CD BHK-21 Production Medium supports suspension BHK-21 cells longer than GMEM supported both by TPB as well as 10% FBS. Cell viabilities started to decline after 24 hours in GMEM, whereas they were stable for four days in CD BHK-21 Production Medium.
Achieve high peak cell densities:
Over four days of culture suspension BHK-21 cells achieve well over two times the densities achieved with GMEM supplemented with TPB (Tryptose Phosphate Broth) and 10% FBS.
Viral titration results in VERO cells
Viral production in triplicate plates using VERO cells grown in either Gibco VP-SFM or EMEM with 10% FBS shows equivalent results
Production of infectious bovine rhinotracheitis (IBR) virus in MDCK cells
The titer of the inoculum was 1.3 x 108 TCID50/mL. Cultures were carried for 6 subcultures in the respective medium. Cell counts shown are average counts (x 106) per flask. The length of incubation was dictated by % cytopathic effects (CPE) in serum control. The two media gave equivalent results.

An economical solution to reduce serum is to replace it with a low serum-requiring medium, supplemented with other growth-promoting factors. Premium Gibco Opti-MEM, a Minimum Essential Medium (MEM) which is supplemented with 2%-4% FBS, actually outperforms ordinary media supplemented with 10% FBS in many critical applications. In some applications, researchers reduced serum requirements by at least 50%. Reducing serum also minimizes the risk of adventitious agents responsible for diseases, such as bovine spongiform encephalopathy (BSE).
Opti-MEM I medium outperforms EMEM with more FBS
For very low serum supplementation (<1%) with anchorage-dependent cells, Opti-MEM I medium should be supplemented with 500 to 1,000 mg/L calcium chloride. The addition of HEPES provides added pH control to the cultures.
