Gibco CHO Media offer high performing options in multiple formats designed to optimize your workflow. We have extended our portfolio to include our newest media formulations: Gibco Efficient-Pro™ Medium and Gibco Efficient-Pro™ Feeds 1 and 2 designed for use with a range of CHO cells. This complete collection provides a range of options for optimal CHO cell culture including High-Intensity Perfusion (HIP) CHO Medium, and Gibco ExpiCHO Stable Production Medium (SPM).
Gibco Efficient-Pro Medium and Gibco Efficient-Pro Feeds 1 and 2 help deliver superior product performance, improved product titer, better protein quality and lot-to-lot consistency, which provides more control and predictability. Each medium is chemically defined, completely scalable and available as Gibco Advanced Granulation Technology (AGT) formats for sustained cell viability. The AGT dry media format enables a simple and scalable reconstitution process. These formats help maximize process efficiency with simplified preparation. Customers with larger-scale operations will benefit from longer bioreactor runs, reduced risk in reconstitution and flexibility to support these CHO cell lines.
Gibco media and process liquids and buffers are packaged in the same high-quality film used for Thermo Scientific bioprocess bags and tubing assemblies.
CHO cells cultured in Efficient-Pro media demonstrated improved protein production—High titers established across CHO variants and vessel size show increased end-product yield, viable cell density, reduced batch numbers, and lower cost with CHO-K1 and CHO-S cells.
CHO-K1 Cells
Efficient-Pro Feed 1 showed comparable productivity to the competitor. Efficient-Pro Medium and Feed 1 maintained high viability and lower cell density indicating a higher specific productivity compared to the competitor.
Glycan and charge variant profiles—Improved protein quality established across all CHO variants creating increased end-product yield, reduced purification steps, and improved efficacy in CHO-K1 and CHO-S cells.
CHO-K1 charge variant profile
Charge variant profile of CHO-K1 cells. Use of Efficient-Pro Medium and Feed 1 resulted in a preferred charge variant profile with a more neutral profile than competitor medium and feed.
High specific productivity of IgG across nearly all CHO variants and vessel size demonstrates opportunity for improved yield, enhanced protein production, and extended harvest stage in CHO-K1, CHO-S, and DG-44 cells.

Gibco High-Intensity Perfusion (HIP) CHO Medium can be used at multiple concentrations to support your process from seed train to production.
Just as it can be used at multiple concentrations, it can be used in multiple perfusion processes: N-1 and seed train, intensified fed-batch, concentrated fed batch, and continuous perfusion. Numerous CHO cell lines (CHO-S, CHO-DG44, CHO-GS, CHO-K1) have been tested in these perfusion processes with HIP CHO Medium.
In addition, the medium supports high cell densities at low medium exchange rates, which improves process efficiency, as low as 1 vessel volume/day (VVD).

There are several pathways to find the right media or media and feed solution for CHO cell cultures. These pathways include selecting a standard or off-the-shelf media, our CHO media test panel, or a media development or optimization service. In order to find the right path, we need to remove the biggest obstacle; luck.
Often time, your goals on what you want to accomplish will narrow down the best selections for you.
Questions to consider:
Comparison of the different options available

Time | Fast | Relative speed | Dependent on scope; >3 months |
Budget | Economical | Mostly cost-effective | More investment needed |
Titer | Average to meets requirements | Meets requirements | Meets or surpasses requirements |

Easy scale-up Liquid and dry formats available that provide performance consistency and may reduce workflow steps to simplify your process. Risk mitigation and assurance of supply May be produced at either of our harmonized manufacturing locations. Available data Frequently, used in application notes and further testing. | Capacity Secondary sourcing options; easy scale-up More specificity Options that are more specific to your clone and molecule. Relative speed Speed is important but not as much as matching product quality. Regulatory support For the panels, provides regularity support needed to succeed in getting approvals. | Resources and/or expertise Need to outsource or consultation on your process Secondary sourcing Increase capacity of your own media and mitigate risk Scale-up solutions Transitioning from a liquid to a dry format during scale-up or performance support during scale-up or tech transfer Improved performance Optimal solution that delivers on your performance criteria Platform media Options that work better than current platform for multiple molecules Regulatory support We offer royalty-free CHO cell lines and documentation for regulatory filings. |
Frequently, cell line specific component dependencies, like insulin and other growth factors, are missed when evaluating media. These are components that are needed for your cell line to thrive (growth, productivity) and if they are not present in the tested media, you may miss out on the optimal solution.
Matching media to your cell line should include an evaluation of whether these dependencies are in the media of choice or not (Tables 1 and 2) to determine whether you need to add them in or not.
Even with the cell line dependency component, in most cases, adaptation is required to see optimal results.
Additional components may also need to be considered for an optimal outcome (Table 3).
Questions:
