Biotinylated reagents for improved protein binding
Biotinylated reagents including biotin, biotin-X, and biotin-XX that are conjugated to secondary antibodies can be used to enhance subsequent binding to proteins such as streptavidin and avidin. Products that are denoted with an ‘X’ represent those that have a seven-atom aminohexanoyl spacer between biotin and the reactive carboxylic acid. This spacer helps to enhance binding to the biotin-binding pockets of avidin and streptavidin. DSB-X Biotin reagent also utilizes a seven-atom spacer to increase binding to streptavidin, avidin, or other biotin-binding proteins. However, DSB-X biotin conjugates can be reversibly bound to streptavidin or avidin, leaving the molecule of interest unchanged.

Tyramide SuperBoost technology for use with biotin labeled secondary antibodies
Tyramide SuperBoost Signal Amplification is a highly sensitive method for the detection of low-abundance targets in multiplexable fluorescent IHC/ICC/ISH experiments. SuperBoost kits combine the brightness of Alexa Fluor dyes with the enhancement of tyramide signal amplification to produce a superior signal. For use with biotin conjugated antibodies use one of the Alexa Fluor Tyramide SuperBoost kits with streptavidin in table X below.

Amplify the signal of your biotin labeled antibody using the Biotin XX Tyramide SuperBoost Kit, Streptavidin (Cat. No. B40931). This specific kit contains biotin XX tyramide with HRP-conjugated streptavidin, which is used prior to detection and signal amplification with an Alexa Fluor-conjugated streptavidin molecule. Figure 1. ATP Synthase detection with Biotin XX Tyramide SuperBoost Kit. HeLa cells were fixed and permeabilized with Image-iT Fixation/Permeabilization Kit (Cat. No R37602). For ATP synthase detection, cells were labeled with anti-ATP Synthase primary antibody and a biotin-conjugated secondary antibody. Biotin signal was enhanced using Biotin XX Tyramide SuperBoost Kit – Streptavidin (Cat. No B40931) and detected with Alexa Fluor 488 Streptavidin (Cat. No. S11223). For best antifade protection, cover slip was mounted with ProLong Diamond Antifade Mountant (Cat. No. P36961).
