Publication-quality imaging

The CellInsight family of high-content screening (HCS)/high-content analysis (HCA) platforms are designed to provide automated imaging versatility and accelerated throughput for the fastest time-to-data possible. Choose from widefield, confocal, or brightfield automated imaging modes, all within one protocol. Our instruments provide up to 12 colors to choose from, from ultraviolet through near-infrared fluorescence and multicolor brightfield imaging to enable flexible imaging paradigms. Finally, our systems are engineered with speed in mind, enabling real-time imaging and analysis conducted in parallel for the fastest time-to-data possible.

Breast cancer tissue cells

CellInsight CX7 LED Pro. Human breast cancer histology slides were stained with H&E and KI67-associated DAB markers (imaged with red, green, and blue transmitted light respectively and a 20X objective lens and the 3-channel Histology bioapplication.

Onboard real-time analysis software

The Thermo Scientific HCS Studio Cell Analysis Software expertly balances flexibility and ease of use to enable meaningful data analysis from single cells through populations. Parallel imaging and analysis offers true real-time results, fast. 

 

HCS Studio software allows backtracking of each individual cell or cellular event to minimize artifacts in the data. View the corresponding cell level cutout associated with each data point to ensure quality control of what you are actually measuring. 

 

To accelerate assay development, every parameter can be evaluated for assay performance based on Z-prime statistical analysis. This allows you to select your top-performing assays as you scale your screening campaigns. HCS Studio software also features guided workflows with user-friendly visual feedback to facilitate easy assay protocol setup, image acquisition, analysis, and scanning.

 

The HCS Studio software for CellInsight CX7 LED Pro and CellInsight CX7 LZR Pro also feature the Thermo Scientific EurekaScan Finder software. This software significantly helps reduce your instrument runtime and analysis efforts by automating the detection and subsequent capture of events at higher magnification.

 

EurekaScan Finder software offers two different approaches based on the type of sample and user-stipulated criteria that will inform high-content screening.

Scenario I: when studying tissues

The selected objects in the first pass are bigger than the second pass’s field of view (case A), or multiple objects appear in the second pass’s field of view (case B). In this case, the second pass uses a uniformly spaced lattice pattern to define the second pass’s field locations.

Scenario II: when studying spheroids, rare-events

The selected objects in the first pass are smaller than the second pass’s field of view and are spaced apart such that multiple objects are unlikely to be in the same field of view. In this case, the second pass centers the fields of view around each object.

Cell Painting multiparameter phenotyping using the Invitrogen Image-iT™ Cell Painting Kit and the CellInsight CX7 LZR Pro platform.

U2OS cells were either untreated or exposed to various concentrations of niclosamide (0–150 μM), gambogic acid (0–2 μM), staurosporine (0–0.5 μM), or amsacrine (0–100 μM) for 24 hours within a 96-well optic thin-bottom plate. Cells were then labeled with Hoechst 33342 dye (Cat. No. H21486), Invitrogen™ Alexa Fluor™ 488 Concanavalin A (Cat. No. C11252), Invitrogen™ SYTO™ 14 Green Fluorescent Nucleic Acid Stain (Cat. No. S7576), Invitrogen™ Alexa Fluor™ 555 Wheat Germ Agglutinin (Cat. No. W32464), Invitrogen™ Alexa Fluor™ 568 Phalloidin (Cat. No. A12380), and Invitrogen™ MitoTracker™ Deep Red FM dye (Cat. No. M46753). After Cell Painting staining, images were acquired with the CellInsight CX7 LZR Pro platform using the 20x 0.8 NA Olympus X Line objective lens and analyzed using the Cell Painting bioapplication in the HCS Studio 5.0 software.

The CX7 LZR Pro HCS platform is equipped with a laser-based autofocus and a powerful sCMOS camera to boost your experiment's assay window and reduce exposure time, providing amazing acquisition speed. Assay speed is further enhanced with true on‑the-fly phenotyping that collapses analysis and image capture into one parallel stream, allowing you to get your data faster. Additionally, the new Thermo Scientific Image-iT™ Cell Painting Kit has been codeveloped with the CellInsight CX7 LZR Pro platform to enable turnkey multiparameter labeling, imaging, and analysis.

 

All these advanced features—paired with a dedicated, all-new, cell-painting bio-application in Thermo Scientific HCS Studio Software and optional Thermo Scientific Amira Software allow the CellInsight CX7 LZR Pro HCS Platform to simplify your most challenging assays, such as cell painting, morphological tracing in 3D, and immune cell to tumor colocalizations.


Store image and database management software

Thermo Scientific Store™ Image and Database Management Software is a SQL™ or Oracle™ relational database used for storing and managing the images and data automatically transferred from an HCS instrument or imported from another imaging source. It allows for rapid, highly scalable, global access to both data and images. Use the Store Image and Database Management Software after you have collected data and images.
