Glycosylation is a critical quality parameter that is examined closely in regulatory filings. Companies must show a range of control over the pattern they get from their bioreactor runs for consistency of process and performance of the protein being produced. In the case of companies making a biosimilar, they must match an innovator molecule glycosylation pattern closely. For others, it may be a matter of showing consistency and perhaps targeting a profile that may make pharmacodynamics parameters of the molecule more desirable.
Until recent years, control was dictated by the cells used and clones generated, necessitating screening of many clones and/or time-consuming development of cells that had been engineered to express specific glycoforms. More recently, there are published components known to influence glycosylation attributes in cell culture that customers have used to control the glycosylation patterns achieved in some cases. Additionally, there are process parameters that can influence other aspects of process quality that are being studied.
GlycanTune Total Feeds provide components needed to move glycan patterns from mostly G0F to G1F and G2F. This is based on the inclusion timing during the feeding process. The products also contain standard feeding supplement components so that they can be used as a substitute for other feeding when the time is right to gain the glycosylation shift desired.
These Total Feeds can be switched to during a feeding schedule at the point in time that provides the desired shift in glycan profile. That can be determined in a simple experiment that is outlined in a manual provided with testing kits that can help a customer determine the best timing to introduce GlycanTune Total Feeds as part of a daily feeding schedule or less frequent feeding schedule.
GlycanTune Total Feeds were developed for use up to production level scales in the thousands of liters of bioreactor process.
GlycanTune Total Feeds are complete feeding supplements patterned after EfficientFeed A+, B+ and C+ supplements that can be super concentrated and reconstituted by just adding water (no pH adjustments required) to deliver up to 200g/L amounts of nutrients cells may need to actually increase titers. Super concentrating to 2X or 3X will allow for the possibility of adding more feed into the bioreactor without diluting the final product and while utilizing available bioreactor space.
