Beta tubulin is a 55 kDa protein that forms a heterodimer with alpha tubulin to make tubulin, the primary component of microtubules. Microtubules mediate many cell movements, including intracellular transport, ciliary and flagellar beating, chromosome alignment in mitosis and meiosis, and structural support of the cytoskeleton. As beta tubulin is ubiquitously expressed in eukaryotic cells, it is often used as a loading control. Quality Invitrogen beta tubulin antibodies are available for a variety of research needs. Immunofluorescent analysis of beta-3 Tubulin (green) and PAX6 (red) in human iPSC-derived forebrain organoids derived at Day 40. The organoids were fixed with 4% PFA for 1 hour at room temperature, followed by incubation with 30% sucrose solution overnight at 4°C. The organoids were then embedded in OCT and cryosectioned at 5 µm, permeabilized with 0.2% Triton X-100 for 20 minutes, and blocked with 10% donkey serum in PBS for 30 minutes at room temperature. Organoid slices were stained with a beta-3 Tubulin Monoclonal Antibody (2G10) (green; Cat. No. MA1-118) at a dilution of 1:500, and a Rabbit PAX6 polyclonal antibody (red; Cat. No. 42-6600) at a dilution of 1:500 in blocking buffer overnight at 4°C, and then incubated with Donkey anti-Mouse Alexa Fluor 488 (Cat. No. R37114) and Donkey anti-Rabbit Alexa Fluor 568 (Cat. No. A10042) at a dilution of 1:1000 as well as DAPI (blue; 1:25000) in blocking solution at room temperature for 1 hour. Images were taken at 20X magnification. Scale bar: 50 µm.
Immunohistochemistry analysis of formalin-fixed paraffin-embedded human kidney tubules stained using a beta Tubulin polyclonal antibody (Cat. No. PA1-41331) at 10 µg/mL. Staining of formalin-fixed tissues is enhanced by boiling tissue sections in 10 mM sodium citrate buffer, pH 6.0 for 10-20 minutes followed by cooling at RT for 20 minutes.
Flow cytometry analysis of Beta Tubulin in CEM cells (green) compared to an isotype control (blue). Cells were harvested, adjusted to a concentration of 1-5x106 cells/mL, fixed with 2% paraformaldehyde and washed with PBS. Cells were blocked with a 2% solution of BSA-PBS for 30 min at room temperature and incubated with a beta Tubulin Loading Control Monoclonal Antibody (BT&R) (Cat. No. MA5-16308) at a dilution of 1 µg/test for 40 minutes at room temperature. Cells were then incubated for 40 minutes at room temperature in the dark using a Dylight 488-conjugated secondary antibody and re-suspended in PBS for FACS analysis.
