The STEMPRO® Adipogenesis Differentiation Kit supports adipogenesis from bone marrow–derived hMSCs (Figures 1A and 1B), human adipose-derived stem cells (ADSCs) (Figure 1C), and hMSCs derived from human embryonic stem cells.
Figure 1. Adipogenesis-induced differentiation using the STEMPRO® Adipogenesis Differentiation Kit.
A. Bone marrow–derived hMSCs induced under adipogenic conditions for 14 days, fixed and stained for oil red O, a marker for lipid-rich vesicles.
B. Bone marrow derived hMSCs induced under adipogenic conditions for 7 days, fixed and lipid vesicles visualized with LipidTOX Green neutral lipid stain (green). Hoechst 33342 applied as a nuclear counterstain (blue).
C. Human ADSCs induced under adipogenic conditions for 12 days produce lipid containing adipocytes as demonstrated by oil red O staining (200X magnification). Data provided by K. Hicok, Cytori Therapeutics.
The STEMPRO® Osteogenesis Differentiation Kit supports osteogenesis from bone marrow–derived hMSCs (Figures 2A and 2B), human adipose-derived stem cells (ADSCs) (Figure 2C), and hMSCs derived from human embryonic stem cells.
Figure 2. Osteogenesis-induced differentiation using the STEMPRO® Osteogenesis Differentiation Kit.
A. Bone marrow–derived hMSCs induced under osteogenic conditions for 14 days, fixed and stained for alkaline phosphatase (a marker for proliferating osteoblasts).
B. Bone marrow derived hMSCs induced under osteogenic conditions for 28 days, fixed and stained for Alizarin Red S (a dye that specifically binds to calcium matrix formations).
C. Human ADSCs induced under osteogenic conditions for 28 days produce calcium-containing mineralized matrix as demonstrated by alizarin red dye incorporation (200X magnification). Data provided by K. Hicok, Cytori Therapeutics.
