In this application note, we take advantage of the OMIP-009 publication, which is optimized for characterizing the immunological response of human T cells, applying it to the 4-laser, 16-parameter configuration of the Invitrogen Attune NxT Flow Cytometer with Autosampler. Additionally, we discuss the importance of antibody titration and optimizing antibody concentration in panel testing. Furthermore, we review the importance of fluorescence minus one (FMO) controls.
This application note demonstrates how to set up a multicolor flow cytometry experiment so that optimal dyes are selected. Additionally, the criteria for setting up a no compensation panel are described.
