준세포 분획 개요

준세포 분획 절차 개요도. 세포 구획을 세포질 추출 버퍼(cytoplasmic extraction buffer, CEB), 그 이후 막 추출 버퍼(membrane extraction buffer, MEB) 및 핵추출 버퍼(nuclear extraction buffer, NEB)와 순차적 세포 배양을 통해 추출했습니다. Pellet 추출 버퍼(PEB)를 가하기 전 NEB 에 micrococcal nuclease(MNase)를 가하여 세포 pellet으로부터 크로마틴 결합 단백질을 추출하여 세포골격 단백질을 용해하였습니다.

서로다른 마우스 조직으로 제조한 세포질 및 핵 추출물의 총 단백질 프로파일. Swiss Webster 마우스 조직(40 mg)을 수확하여 PBS로 헹군 후 NE-PER 핵 및 세포질 추출 시약 키트로 용해시켰습니다. 추출물을 Pierce 660nm 단백질 분석 시약(Part No. 22660)으로 정량했습니다. 값은 두 개의 별도 분리 값의 평균입니다.
