Horseradish peroxidase (HRP) is a commonly used enzyme label with secondary antibodies to amplify the signal of detected targets. HRP catalyzes the oxidation of substrates in the presence of hydrogen peroxide and the result is either a colored precipitate or light. When chromogenic substrates come into contact with the enzyme a colored product precipitates which doesn’t require additional equipment to visualize. When a chemiluminescent substrate comes into contact with HRP light is emitted as a byproduct and must be captured by an imaging instrument.
Horseradish peroxidase (HRP)-conjugated secondary antibodies are commonly used for chemiluminescent, colorimetric, or fluorescent detection in applications, such as western blotting, ELISA, immuno-histochemistry, and others. For each application, HRP-conjugated secondary antibodies need substrates that can deliver high signal-to-noise ratios for the chosen detection method.

F(ab’)2 fragments result from digestion of a whole antibody with pepsin. These antibody fragments contain the strong divalent binding found in the variable regions but lack the Fc portion of the antibody.
Chemiluminescent detection offers exceptional sensitivity and the ability to reprobe western blots.
Chromogenic substrates offer simple and cost-effect western blot detection.
Invitrogen SuperBoost tyramide signal amplification kits and reagents offer exceptional signal amplification for fluorescent imaging of low-abundance targets. SuperBoost EverRed and EverBlue substrates provide permanent colorimetric staining that is also fluorescent.
