Mass spectrometry can detect very low analyte concentrations in complex mixtures. It can be quantitative with the use of peptide labels or standards that are concomitantly analyzed with the sample and act as a reference point for both relative or absolute analyte quantitation, respectively. We offer three different systems that can be applied to solve a wide variety of biological problems. Relative quantitation strategies include tandem mass tagging (TMT) and stable isotope labeling using amino acids in cell culture (SILAC). For both relative and absolute quantitation, SureQuant assays are designed for multiplexed targeted quantitation for key cellular signaling proteins.
Tandem mass tag (TMT) reagents are robust and efficient tools for multiplex relative protein quantitation of up to 18 samples derived from cells, tissues or biological fluids. All mass tagging reagents within a label reagent set have the same nominal mass (i.e., are isobaric) and chemical structure composed of a reactive group, a spacer arm (mass normalizer), and a mass reporter. The isobaric nature of the TMT reagents enables concurrent identification and quantification of proteins in different samples using tandem mass spectrometry (MS).

The SureQuant assays enable multiplexed targeted quantitation for key cell signaling proteins and contain reagents for multiplexed immunoprecipitation and mass spectrometry sample preparation, heavy peptide quantitation panels, and suitability standards to assess system performance. Together, the SureQuant assay system provides flexibility to use the immunoprecipitation sample prep kits and peptide quantitation modules together or independently based on the end application for robust quantitation.

SILAC metabolic labeling relies on in vivo incorporation of “heavy” 13C- or 15N-labeled amino acids into proteins during translation for multiplexed quantitative comparison of “heavy” and “light” protein. It can be widely applied for comprehensive identification, characterization and quantitation of proteins. We offer a broad portfolio of liquid and powdered SILAC media and SILAC amino acids. These reagents are available as kits or as stand-alone reagents.
